Thanks mate.
I work on a 24hd cintiq hooked up to an iMac running photoshop.
I tend to use about 4 layers.
One for layout/design
One for forms
One to tighten up/ 'pencil'
And one for final 'inks'.

Very cool, I have a similar set-up (22Hd instead) on a Mac as well.

What tools within PS (I'm assuming) do you use for straight lines? I've tried the line tool which is good, except the square corners sometimes, and the point shift click thing is a pain.

Honestly, Im pretty basic with the tools I use, I actually draw with a plastic ruler on my cintiq, I also use a lot of elipse stencils etc.
I found working with a line tool was too neat and controlled for me.

I just got CS6, really like it!
